This mouse IgG2a kappa is purified using salt fractionation procedures and Protein A chromatography. It is produced from tissue culture media obtained at our facility in The United States. Antibody concentration was determined using an absorbance at 280 nm: 1.4 equals 1.0 mg of IgG. This antibody reacts with human IgE (epsilon chain specific) as determined by ELISA and western blot. It is suitable for use in blotting assays, ELISA and lateral flow. Optimal working dilutions should be determined experimentally by the investigator.

The Ig fraction was enriched from normal mouse serum using a combination of procedures that include salt fractionation. Antibody concentration was determined by extinction coefficient: O.D. 1.4 at 280nM equals 1.0 mg of IgG. Estimated purity of >80% as determined by SDSPAGE. Optimal working dilutions should be determined experimentally by the investigator.

Goats were immunized with highly purified mouse IgG1 and the resulting antiserum was collected. Antibody was solid phase adsorbed using mouse IgG2a, IgG2b and IgG3 immunosorbents to remove cross reactive antibody. Specific antibody was immunoaffinity purified off a mouse IgG1 containing immunosorbent. Antibody concentration was determined using an absorbance at 280 nm: 1.4 equals 1.0 mg of IgG. This antibody will react with mouse IgG1 as determined by ELISA and IEP techniques and has less than 1% cross reactivity with mouse IgG2a, IgG2b and IgG3. It is suitable for blotting, ELISA and IHC applications. This antibody may cross react with IgG from other species. Optimal working dilutions should be determined experimentally by the investigator.

Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In non-muscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a muscle-specific, alpha actinin isoform that is expressed in both skeletal and cardiac muscles. MerTK, also called c-Mer, is a member of the Mer/Axl/Tyro3 receptor kinase family. It is a 984 residue transmembrane protein made up of one tyrosine kinase domain, two Fibronectin type-III domains and two immunoglobulinlike C2-type domains. MerTK is the mammalian ortholog of the chicken retroviral oncogene product v-Eyk. This protein plays a critical role in macrophage activation, platelet aggregation, clot stability and the efficient removal of apoptotic cells. Specifically, MerTK acts as a signaling molecule, triggering outer segment ingestion in the retinal pigment epithelium (RPE) phagocytic process. Evidence suggests that MerTK signals via interaction with phosphatidylinositol-specific phospholipase C 2). Recognizes a protein of 55-62 kDa, identified as cyclin B1. In mammals, cyclin B associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at aa 14Thr and 15Tyr. This maintains the inactive state until the end of G2-phase. The inactive cyclin B-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates aa 14Thr and 15Tyr of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when cyclin B is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. So, cyclin B-p34cdc2 plays a critical role in G2 to M transition. Recognizes a protein of ~35 kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas.
